Combate Americas' first open tryout took place in August 2016 in Pasadena, California. The tryout was sponsored by the US Army, and was open to all men and women in all weight classes. The event drew over 200 hopeful fighters.

Former UFC star Danny "Last Call" Castillo hosted the tryout, and among the judges were UFC women's superstar and Combate Americas color commentator Julianna "The Venezuelan Vixen" Pena and referee Mike Beltran.
