The Janitor you play as will always walk forward, so the only key you'll have to worry about is pressing and holding down the left mouse button to charge your swing to full and hit/clean a petpet to stop them from tracking dirt around. Always have your mop charged and ready to clean a petpet, so always be holding the left mouse key. Wait until the petpet is right next to you with a fully charged swing and let it go! 
If there are two petpets close enough that one is directly behind you and one is directly in front of you, you can clean them both at the same time if you've charged your swing and release it.
You can also try to mash the left mouse button and swing the mop a few more times after the first if you can't charge your swing meter fast enough. This way, you can get the petpet just a bit cleaner and prevent your dirt meter from rising too quickly.
If you swing and miss a petpet, the Janitor will get angry and throw a temper tantrum for a few precious seconds, which will freeze you in place and you can't move. To prevent this, you can start recharging the mop before it hits the floor.
As you clean petpets, they will sometimes drop upgrades. You can't avoid them since the Janitor keeps moving forward without your input, but luckily they are all good for you! They upgrade your cleaning Power, will give you Honey to decrease your temper tantrum time, Money gives you points, Speed doubles the janitor's walking and power rejuvenating speed, making smacking petpets easy.
Along the level you'll collect sparkling tears of water, which fills up your water bucket. When it's full, you can press space to throw the water on all the petpets in front of you. Use this to clean a crowd of petpets that are close together, preferably.
Every time you pass King Aldator (completed a lap), you'll be able to upgrade your mop handle or mop head. 

For mop head, for level 2 you'll have to either sacrifice cleaning power (how clean you make a petpet in one swing) for a bigger cleaning window (how long is your reach/range with the swing). For level 3, middle option is the balance between the two, left option is best cleaning power but small cleaning window, right option is large cleaning window but okay cleaning power.
For mop handle, for level 2, you'll have to either sacrifice walking power (movement speed) for a faster power up speed (how fast you can charge your swing to max power). For level 3, the middle option is the balance between the two, left option is best walking speed but slow power up speed, right option best power up speed but slow walking speed.

Petpets (from Jellyneo Guide):
-Minitheus is the most common and simplest petpet. It walks along at a fair pace, easy to hit until they start coming in multiples with other types of petpets.
-Altachucks are basically just like Minitheuses, though a bit faster; they're still nothing much to worry about.
-Alabrisses, too, are fairly easy to clean and aren't too fast, though there's one unimportant detail different from the first two petpets: they fly. Only a foot or two off of the ground, fortunately, but at least it's something different.
-Vaeoluses, the game starts getting difficult. They also fly, but they're much quicker than Alabrisses. When they start appearing, you've got to be more alert.
-Altalaphuses bounce, and they're possibly the hardest petpet to clean because of their speed; if you're not careful, you might miss some of them entirely.
