3D rendering of a mouse intestinal crypt obtained with a two-photon scanning microscopy. The image cuts away from the apex of the villi towards the crypt's base to reveal the intestinal stem cells (green), expressing prominin 1 and yellow fluorescent protein (YFP). Prominin 1 is a glycoprotein often expressed on the membrane of adult stem cells, where it helps to maintain stem cell properties by suppressing differentiation. Method: A one-millimeter slice of intestine from a Prom1+/C-L; Rosa26-YFP; Ctnnb1+/lox(ex3) mouse was placed on a glass cover slip and submerged in Kreb's solution superfused with a gas mixture of 95% O2/CO2. Microscopy was performed with an Ultima imaging system (Prairie Technologies) and a Ti:sapphire Chameleon Ultra femtosecond-pulsed laser (910 nm) (Coherent).
