Live zebrafish embryo expressing GCaMP5. Image of the spine was taken with 2P excitation at 920 nm, 9 z planes over 18 microns. Customer description: I study early spontaneous activity and intrinsic photosensitivity in the embryonic zebrafish spinal cord. With two-photon calcium imaging, we can observe neural activity and reserve the visible spectrum for targeted photostimulation of endogenous photoreceptors. At these early ages, the spinal cord is curved and it is impossible to get a single optical section containing all active cell types—2p volumetric imaging lets us sample activity in cell types at different positions along the dorsal-ventral axis, while still imaging from both the left and right sides of the spinal cord. Further, with Airsycan fast mode, the observation of neurons firing at different depths of the spinal cord is possible.
